GLP-1 receptor agonists (GLP-1 RAs) and dual GLP-1/GIP agonists, which are all peptide-based medicines, are important treatments for type 2 diabetes and obesity. Most of these drugs, such as semaglutide and liraglutide, are administered by injection. The first oral option, semaglutide (Rybelsus™), suffers from very low bioavailability, with only about 1% of the dose absorbed, partly due to the gastro-intestinal mucus barrier. This project aims to develop alginate oligosaccharides (AOs) as innovative functional excipients to enhance mucus permeability and significantly improve the oral uptake of GLP-1 RAs and dual GLP-1/GIP agonist drugs. The anticipated benefits include better patient adherence, reduced side effects, and the establishment of a platform technology with potential applications to other biologic medicines.
